大鼠浅Ⅱ度烫伤创面EGF及受体基因的表达 被引量:6

Expression in Epidermal Growth Factor and Its Receptor Gene During the Partial Thickness Burn wound Healing of Rats

摘要 取大鼠浅Ⅱ度烫伤后1、3、5、7、10d和14d及正常对照(脱毛未烫伤)的皮肤样本,采用核酸分子斑点杂交技术,观察表皮生长因子(EGF)及其受体(EGF-R)的基因表达。结果发现:EGF基因表达水平于伤后第3d明显增强,第5d达高峰;EGF受体基因表达水平于伤后第1d即明显增强,伤后第5d恢复正常。提示内源性EGF及其受体参与了调控创面修复,因子和受体之间相互制约,避免细胞过度增生。 Epidermal growth factor (EGF) has been shown to stimulate the proliferation and differentiation of keratinocytes and enhance wound healing. EGF acts with the EGF receptor (EGF-R). In order to demonstrate the expression in EGF and EGF-R gene in tissue during the partial-thickness burn wound healing of rats, 48 SD rats were divided into 7 groups. Wound surface of 1o% TSBA were made on the back of every rat by 70℃ thermal water, acting for 8 seconds. After 1 day and 3,5,7,10and 14days, the skin samples from wound surface of one group of rats and from normal control were collected. EGF and EGF-R gene mRNA were detected by slot blotting hybridization technique. The results showed that EGF gene expression increased significantly after 3days, and reached to peak after 5 days. EGF-R gene expression increased significantly after 1 day, and reached to normal after 5 days postburn. It suggested that EGF and EGF-R are involved in the regulation of wund healing. EGF and EGF-R have a regulation effect on each other to avoid cell overgrowth.
