生长因子对培养肝贮脂细胞增殖与游走的刺激作用

摘要 从大鼠肝分离贮脂细胞,培养5d后,分别添加碱性纤维母细胞生长因子(bFGF)、肝细胞生长因子(HGF),观察其对贮脂细胞增殖与游走的影响。结果,对照组贮脂细胞的BrdU标识率为18%,HGF组为17%,bFGF(50 mg/L)组达58%(P<0.01);细胞的游走速度,对照组为6.4μm/h,HGF组为6.5μm/h,bFGF组为16.6μm/h(P<0.01)。显示bFGF能促进贮脂细胞的增殖和游走,但HGF无类似作用。提示肝炎、肝纤维化时,病变局部贮脂细胞的大量积集可能与纤维母细胞生长因子有关。 Fat-storing cells are accumulated in the lesion of liver fibrosis in clinical and experimental con- ditions. However, the mechanism in detail of fat-storing cell accumulation is still unknown. In this study, we examined the effects of basic fibroblast growth factor (bFGF) and hepatocyte growth factor (HGF) on fat- storing cell proliferation and migration using primary cultured rat fat-storing cells. Fat-storing cells were pre- pared from wistar strain rats and were cultured in DMEM with 10% fetal calf serum for 5 days. Proliferation and migration speed were assessed with or without growth factor using BrdU staining and time-lapse VTR. The BrdU labeling index was 18% in the controls, 17% in the HGF group and 58% in the bFGF group (P< 0.01). The migration speed was 6. 4μm/h in controls, 6.5μm/h in the HGF group and 16.6μm/h in the bFGF group (P<0.01). The proliferation and migration were significantly accelerated by bGFG but not by HGF. In conclusion, bFGF might play a role in the accumulation of fat-storing cells in the lesion of liver fi- brosis.
