肾灌注、环孢霉素A对豚鼠至大鼠异种移植心存活时间的影响

摘要 目的:本研究旨在观察肾灌注、环孢霉素A对豚鼠至大鼠异种移植心存活时间的影响。方法:采用肾灌注吸收大鼠体内天然抗体、环孢霉素A进行免疫抑制,异种心脏移植应用异位颈部移植方法。结果:单纯应用环孢霉素A,移植心存活时间为(13.6±4.6)min;单纯应用肾灌注,移植心存活时间为(161.9±36.3)min(P<0.01);环孢霉素A结合肾灌注移植心存活时间为(385.2±186.5)min(P<0.05),肾灌注后24h作心脏移植,移植心存活时间为(13.8±5.1)min(P>0.05),术前用环孢霉素A,肾灌注后24h作心脏移植,移植心存活时间为(178.3±38.7)min(P<0.01)。结论:单纯应用环孢霉素A不能延长豚鼠移植心存活时间,单纯肾灌注能明显延长移植心存活时间,环孢霉素A结合肾灌注能更进一步延长移植心存活时间。环孢霉素A可能抑制肾灌注后大鼠体内天然抗体水平的早期反跳。 Objective: To observe the effect on cardiac xenografts survival from guineapigs to rats. Methods: The natural antibody in rats was absorbed by guineapig kidney perfusion, while immunosuppression was completed by cyclosporine. The technique of heterotopic cervical cardiac xenotransplantation was applied. Results: With single use of cyclosporine the cardiac xenograft survival time was 13.6±4.6 min, while with sole kidney perfusion it was 161.9±36.3 min (P<0.01). However, cardiac xenograft survival was 385.2±186.5 min (P<0.05) with the above two methods combined. When cardiac transplantation was done on the following day after kidney perfusion, the graft survival time was 13.8±5.1 min (P>0.05). When cyclosporine was applied preoperatively in the previous situation, the graft survival time was 178.3±38.7 min (P<0.01). Conclusion: The guineapig cardiac xenograft survival is not prolonged by single use of cyclosporine, while single kidney perfusion obviously can do. When kidney perfusion is combined with cyclosporine, the survival time is further prolonged. Cyclosporine may inhibit the early rebound of natural antibody after kidney perfusion. 
