摘要
使用硫代修饰的G蛋白(Gαq/11亚基和ras蛋白)反义寡聚脱氧核苷酸(Gαq/11亚基和ras蛋白AS-ODNs)作用于含20%胎牛血清DME培养液培养基的血管平滑肌细胞(vSMC)。通过细胞计数和免疫细胞化学方法,观察了Gαq/11亚基和ras蛋白AS-ODNs对vSMC增殖和PCNA表达的影响。结果表明,Gαq/11亚基和ras蛋白AS-ODNs联合应用能明显抑制vSMC增殖;免疫细胞化学分析亦显示vSMC中PCNA蛋白表达显著减少。而相同浓度的正义ODNs只有微小抑制作用。结果提示,Gαq/11亚基和ras蛋白AS-ODNs有可能应用于防治临床血管活性物质依赖性细胞增生性疾病的研究。
Antisense phosphorothioate oligodeoxynucleotides(AS ODNs) were added to vSMC cultured in 20% fetal calf serum DME media.Using cell count and immunocytochemistry methods,the cell growth and expression of proliferating cell nuclear antigen (PCNA) of vSMC were studied.Results showed that the joint administration of Gαq/11 subunits and small molecular G protein(ras rotein)AS ODNs could significantly inhibit vSMC proliferation.Immunocytochemistry analysis show that expression of PCNA protein in vSMCs also decreased evidently.When Gαq/11 subunits and ras protein sense ODNs were used instead of AS ODNs,only a little effect was demonstrated.In conclusion,Gαq/11 subunits and ras protein AS ONDs may be used for clinical research of prevention and treatment of vasoactive factors dependent proliferating diseases.
