异丙酚对大鼠小脑、海马和皮层环鸟苷酸含量的影响

摘要 研究异丙酚对大鼠脑组织环鸟苷酸(cGMP)含量的影响。方法16只SD大鼠随机分为2组,分别腹腔注射(ip)生理盐水10ml·kg-1(对照组)和异丙酚100mg·kg-1,待翻正反射消失后,立即断头,0~4℃下取脑,分区,匀浆,离心,抽取上清液用4倍体积的水饱和乙醚冲洗4次,然后置60℃水浴上蒸干制备组织样品,用放射免疫法测定不同脑区脑组织cGMP含量。cGMP放射免疫测定使用cGMP测定试剂盒。结果ip异丙酚100mg·kg-1能明显降低大鼠小脑、海马和大脑皮层的cGMP含量(P<0.05或P<0.01)。结论在大鼠异丙酚的全麻作用可能与其减少脑组织cGMP含量有关。 Objective To investigate the effects of propofol on cyclic guanesine 3′,5′-monophosphate (cGMP) content in rat brain.Methods sixteen SD rats were divided randomly into two groups.The aminals were administred introperitonealy (ip) noraml saline 10ml·kg -1 (control group) and propofol 100mg·kg -1 , respectively. These rats were immediately decapitated after the loss of righting reflex.In order to prepare brain tissue samples,brain tissues were dissected on ice, and then gomogenized and centrifuged. The supernatant fluids wee washyed and extracted four times with 4vol of water saturated with diethyl ether. The extracts were dried over 60℃ water bath. cGMP was measured by radioimmunassay in dried extract samples from different braim areas. The radioimmunoassay of cGMP was performed by using cGMP assay kit.Results Propofol 100mg·kg -1 ip significantly raduced the cMGP content in the cerebellum, hippocampus and cerebral cortex as compared with that of control group (P<0.05 or P<0.01).Conclusion The role of propofol in general anesthesia may be related to the reduction of cGMP content in brain tissues of the rat.
