摘要
目的观察FK506对大鼠肾转化生长因子-β(TGF-β)和纤维连结蛋白(Fn)基因表达的影响。方法对FK506组8只大鼠行部分肾切除术以造成与人肾移植相似的模型。以Northern杂交方法,采用鼠TGF-β和Fn基因探针检测大鼠肾TGF-β和Fn基因的表达。结果大鼠肾TGF-β和Fn基因的表达在FK506组明显高于对照组。以光密度分析仪将杂交斑点密度转换为吸收度,发现FK506组TGF-β和Fn基因的表达水平分别是对照组的3.5倍和2倍。结论FK506可以增强大鼠肾TGF-β和Fn基因的表达,促纤维合成细胞因子和糖蛋白在FK506诱导肾毒性中的作用可能与此有关。
Objective To observe the effect of FK506 on expression of transforming growth factor-β (TGF-β) and fibronectin(Fn) genes in rat kidney.Methods Nephrectomy was performed on 8 rats.With rat TGF-β and Fn probe,the expression levels of TGF-β gene and Fn gene in 8 rat kidney were analyzed by Northern blot.Results Compared with control group,the expression levels of TGF-β gene and Fn gene were relatively hygh in FK506 group kidney.By densitometric analysis,350% change was found over control group kidney in TGF-β gene expression and 200% in Fn gene expression.Conclusion The present study demonstrates that FK506 enhances TGF-β and Fn gene expression in rat kidney,which may be associated with the roles of fibrosis-promoting cytokine(TGF-β)and glycoprotein (Fn) in FK506-induced nephrotoxicity in rats.
