摘要
报道RP-HPLC法测定脑得生片中葛根素含量。采用Shim-Pack CLC-ODS(15cm×6mmID)分析柱;北分厂ODS保护柱;流动相:甲醇-水(25:75);流速:1.0ml/min;色谱柱温度:35℃;ShimadzuFLD-6A荧光检测器(激发波长:350±20nm;检测波长450nm-800nm);进样体积10μl;外标法计算含量。理论塔板数按葛根素峰计算为2601,葛根素浓度在20μg/ml-120μg/ml范围与色谱峰面积呈线性关系,r=0.9998,平均回收率为99.46%,RSD1.5%。
A quantitative method was developed for the determination of puerarin in Naodesheng Pian by reversed phase HPLC. Chromatographic conditions included column ODS-C18, column temperature, 35°C ; FLD-6A fluorescence detector (excitation wavelength 350±?20nm;fluorescent wavelength 450nm-800nm) ; mobile phase, methanol-water (25:75),flow rate 1ml/min; injecting volume, 10μ1; external standard. The number of theoretical plates calculated by puerarin peak was no less than 2000. The standard curve was linear in the concentration range of 20μg/ml-120μg/ml, and correlation coefficient was 0.9998. The average recovery and the rel-ative standard deviation were 97.2%and 1.6%, respectivily.
