松果腺褪黑激素对大鼠下丘脑视交叉上核AVP VIP肽能神经细胞的影响 被引量:2

摘要 本研究选用成年健康雄性Wistar大鼠建立松果腺切除和松果腺褪黑激素(MLT)注射模型,并设置正常、松果腺切除假术、生理盐水注射三种对照组。MLT注射时间分别为1000,1700,2400。实验组、对照组动物均存活30天,于上午900麻醉、固定、取材、切片、ABC免疫组化染色、显微图象分析系统(MIAS)相对定量分析处理,目的在于观察MLT对SCN不同肽能神经细胞的作用。结果表明:MLT对SCN内AVP样神经细胞无显著作用,而可促使其VIP样神经细胞中VIP含量增多。 Adult healthy male Wistar rats were divided into experimental and control groups. The experimental groups contain pinealectomy group and melatonin(MLT) injection group. The control groups consist of normal control, saline injection and sham-pinealectomy group. Rats of the MLT injection groups were injected subcutaneously with MLT at 10∶00, 17∶00, 24∶00 daily respectively. All animals were killed after 30 days. They were anesthetized and perfused through heart with fixative which contained 4% paraformaldehyde and 0 26% picrio acid at 9∶00. Sections were cut with freezing microtome, stained with immunohistochemical ABC technique and measured with MIAS for semiquantitative analysis. The investigation aims at studying the influence of MLT on different peptidergic neurons in SCN. The results indicate that the injected MLT has no influence on AVP content but increases VIP content in the SCN neurons.
