摘要
采用免疫电镜方法观察高温情况下胶体金标记的刀豆素A(ConA-Au)与游离培养的大鼠腹腔巨噬细胞表面受体结合、内吞及其在巨噬细胞中转运情况,探讨高温对RME的影响。实验组(A、B、C、D、E)收集大鼠腹腔巨噬细胞与ConA-Au共同在恒温水浴箱中孵育,温度分别为39、40、41、43、45℃,于不同时间间隔(、10、20、30、60、120 min)常规电镜制样;对照组分别为腹腔注射ConA-Au(F)及胶体金(G)。结果表明:随着温度升高RME抑制逐渐增强,45℃时RME完全受到抑制,低于41℃时可见到溶噬现象。
The purpose of this study was to approach the effects of high temperature on receptor-mediated endocytosis in culture cells, observing by immuno-electron microscopy the binding of colloid gold labelled ConA (ConA-Au) with the surface receptor of rat peritoneal macrophages freely cultured, endocytosis and transmission within macrophages at high temperature. In the five experimental groups,the rat peritoneal macrophages were collected and incubated with ConA-Au at different temperatures (39℃~45℃ ), fixed for different intervals (5 min^120 min) and prepared for electron microscopic observation as routine. The two control groups were injected intra-abdominally with ConA-Au and pure colloidal gold solution respectively. The result showed that RME inhibition increased obviously with the rising of temperature and reached the completion at 45℃. The lysosomophagy could happen below 41℃.
