等位基因特异性PCR检测HBV核苷酸1858碱基及与1896G→A变异的关系

IMPLICATION OF NUCLEOTIDE 1858 OF HEPATITIS B VIRUS GENOME FOR NUCLEOTIDE 1896 G TO A MUTATION

探讨乙型肝炎病毒前C区终止变异(nt1896G→A)与核苷酸1858碱基的关系。方法 等位基因特异性PCR检测核苷酸1858碱基,错配PCR柱测核苷酸1896G→A变异。结果 显示58例乙型肝炎患者检出1858-T株感染54例,4例为1856-T/C混合株感染,共检出A1896变异33例(57％);结论 中国人乙型肝炎患者发生前C区A1896变异可能与绝大多数为1858-T株感染有关。

To study the relation of nueleotide (nt) 1858 to mutation of nt 1896 G-A in patients with hepatitis B virus infection, detection of nueleotide 1858 by allele-specific polymerase chain reaction (PCR) and nt 1896 G-A mutation by mispairing PCR were conducted.54 of 58 patients with hepatitis B were detected to be with 1858-T strain and 4 with 1858-T/C mixed strain infection.A1896 mutation rate was 57% (33/58).The results shows that the high positive rate of A1896 mutation in Chinese patients is possibly related to the high 1858-T strain infection.
