小鼠肿瘤疫苗的制备及鉴定

摘要 将小鼠骨髓瘤细胞(SP2/0)与脂多糖(LPS)活化的B淋巴细胞在聚乙二醇(PEG)介导下融合,经筛选获得8株状态良好的阳性细胞。这些细胞(传代第二次)能耐受HAT液,而对8—氮杂鸟嘌呤(20μgml)敏感,其染色体数在105~112条之内,表明这8株细胞均为SP2/0细胞与B细胞融合而成的杂交细胞。其中三株杂交细胞(IF6、IF11、2A6)能显著增强T淋巴细胞的增殖反应(P<0.01),提示IF6、IF11、2A6细胞对T细胞具有共刺激作用。其表面可能表达了B7等共刺激分子。 Eight strains of hybrid tumor cells were established by fusion of SP2/0 myeloma cells with LPS -activated B cells mediated by PEG. The hybrid cells (passage two) were tolerant to HAT, but sensitive to 8-Azaguanine (AZ) (20ug/ml) and their chromosome numbers were from 105 to 112. Three of the hybrid cells (1F6,1F11, 2A6) could markedly enhance proliferation of T lymphocytes treated with PMA, which suggested that the hybridoma cells provideda costimulatory signal to T cells and there were B7 costimulatory molcuies exprssion on their surfaces.
