光辐射烧伤后大鼠PMN吞噬功能变化机理研究

摘要 大鼠15%TBSAⅢ度光辐射烧伤后第7和14d外周血PMN—CL和PMN NADPH氧化酶活性显著降低,铜锌SOD活性和LPO含量显著升高;PMNC_(3b)受体表达于伤后第1、7d显著升高,PMNMPO活性伤后第7d显著降低;伤后PMN—CL降低与PMN NADPH氧化酶和MPO活性降低、铜锌SOD活性升高及LPO含量增加显著相关;提示伤后NADPH氧化和MPO活性降低及SOD活性升高是造成PMN—CL下降的重要原因;而PMN功能损伤可能与脂质过氧化的损伤作用有关。 A rat model of 15% TBSA full thickness thermal burn was employed so as to study the changes,induced by the burn,of polymorphonucler leukocyte chemiluminescence(PMNCL),PMN C3 receptor expression, activity of NADPH oxidase, superoxide dismutase (SOD) and myeloperoxidase (MPO) and lipid peroxide(LPO) level on the 1st ,7th, 14th, 21th and 28th day after the injury. It was found that the intensity of PMN CL and the activity of NADPH oxidase decreased significantly and the activity of NADPH oxidase decreased significantly on the 7th and 14th day;the expression of PMN C3 receptoy increasd on the 1st and 7th day,the activity of PMN MPO decreased singnificantly on the 7th day. There were close correlation between PMN CL and PMN NADPH oxidase activity (r = 0. 865,P< 0. 001),detween PMN CL and SOD activity(r = 0. 8986,P<0. 001) ,between PMN CL and MOP activi-ty(r = 0. 5608,P<0. 005) and between PMN CL and LPO activity (r = -0. 8136,P<0. 001) ,respectively. It was indicated that the changes of PMN NADPH oxidase,MPO and PMN SOD axtivites were the main cause of PMN dysfunction amd the dysfunction might be related to lipid peroxidation damage to PMN.
