葛根素对高脂血症大鼠脂质代谢及血浆PGI_2、TXA_2水平的影响

Effects of puerarin on the levels of PGI_2 and TXA_2 in plasma of rats with hyperlipidemia

目的 观察葛根素对高脂饲料诱导的高脂血症大鼠血浆前列腺素I2 (Prostacyclin ,PGI2 )和血栓素A2 (Throm boxane ,TXA2 )水平的影响 ,为防治高脂血症提供实验依据。方法 48只Wistar大鼠 ,随机分为 4组 ,分别饲以正常饲料、高脂饲料、葛根素干预Ⅰ组和葛根素干预Ⅱ组 ,每组 12只。饲养 8周后测定大鼠血中脂质PGI2 、TXA2 的含量。结果 葛根素能显著抑制高脂饲料诱导的大鼠血浆甘油三酯、总胆固醇、低密度脂蛋白胆固醇、以及血栓素A2 升高、降低TXA2 /PGI2值 ,同时显著升高高密度脂蛋白胆固醇 (F =10 9 163 ,P =0 0 0 0 )。结论 葛根素具有明显的降血脂作用 ,同时能降低TXA2 /PGI2 。

Objective To investigate the effects of puerarin on levels of thromboxane A 2 (TXA 2) and prostaglandin I 2 (PGI 2) in plasma of rats with hyperlipidemia. Methods A total of 48 Wistar juvenile rats were randomly divided into four groups: normal diet, high fat and high cholesterol diet (HFD), HFD supplemented respectively with 1 g/kg puerarin (puerarin group Ⅰ) or 2 g/kg puerarin (puerarin group Ⅱ). After eight weeks of feeding, the rats were sacrificed for detection of the levels of lipid, TXA 2, and PGI 2. Results The blood triglycerol, total cholesterol, LDL C, TXA 2, and TXA 2/PGI 2 were significantly lower in puetarin Ⅰ and puerarin Ⅱ groups than those in HFD group, but the concentration of HDL C was significantly increased in the supplemented group( F =109 163, P =0 000; P <0 05). Conclusion Puerarin can decrease blood lipids and TXA 2/PGI 2 ratio, suggesting that puerarin has the effects on anti platelet aggregation and the prevention of atherosclerosis.
