糖尿病小鼠对流感病毒灭活疫苗的免疫应答

Immune Responses to Influenza Vaccine in Diabetic Kunming Mice

摘要 目的 用不同剂量的流感病毒灭活疫苗免疫糖尿病小鼠 ,检测糖尿病小鼠对流感疫苗的免疫应答。方法 利用一次性大剂量注射链脲佐菌素 (streptozotocin ,STZ)的方法 ,建成昆明小鼠糖尿病模型 ,用流感病毒灭活疫苗免疫正常小鼠和糖尿病小鼠 ,用 3 0、 1 5、 1 0、 0 5和 0 1μg的剂量 ,免疫一次后间隔 3周 ,再加强免疫一次 ,分别取血用ELISA法测免疫一次和免疫两次后的IgG抗体。结果 用 3 0、 1 5、 1 0、 0 5和 0 1μg的剂量免疫后 ,无论是免疫一次还是加强免疫后 ,糖尿病小鼠产生的抗体量均低于正常小鼠产生的抗体量。结论 糖尿病小鼠由于自身免疫机制缺陷 ,对抗原免疫原性反应较正常小鼠差。 Objective Diabetic Kunming mice were immunized with different dose of inactivated influenza (A/PR8)vaccine to examine the immune responses to influenza vaccine. Methods Diabetes was induced in Kunming mice by injection of a single high dose streptozotocin (STZ),then the mice were immunized twice 3 weeks apart with various dosage (3.0μg,1.5μg,1.0μg,0.5μg and 0.1μg)inactivated influenza vaccine.Sera from the mice were analysised for antibody titers by ELISA. Results In mice immunized with 3.0μg,1.5μg,1.0g,0.5μg and 0.1μg inactivated vaccine,the level of antibody titer in diabetes mice group was distinctly less than that of healthy mice group. Conclusions The diabetic mice have a poor immune response to influenza vaccine.
