摘要
建立了食品中雌三醇、雌二醇、雌酮、睾酮、孕酮5种激素同时分离测定的高效液相色谱分析方法。各类食品样品经固一液、液一液萃取处理后,以甲醇一乙腈—四氢呋喃—0.01mol/L醋酸钠溶液为流动相,安眠酮为内标进行反相液相色谱分析。5种激素分离效果良好,各组分浓度1.0~32μg/mL标准曲线线性关系好,回归分析的相关系数均大于0.999。方法的最低检测浓度:雌三醇0.009mg/kg;雌二醇0.038mg/kg;雌酮0.044mg/kg;睾酮0.020mg/kg;孕酮0.071mg/kg。5种激素同时分离测定的相对标准偏差小于15%不同浓度5种激素同时回收的回收率均在80%以上。该方法操作简便、准确、灵敏,满足实际样品测定需要。应用该方法对市场上的部分食品分析的结果表明,一些营养保健食品中会有不同成分的激素,提示有关部门应尽快制定法规进行监督管理。
method for determination of
sex hormones
in food stuff was reported.Samples was extracted with solid-liquid
and liquid一liquid extraction, useing hyminalas internal standard
substance and RP-HPLC separation was performed on C18 ascolumn with
methanol-methyl cyanide-tetrahydrofuran-.01 mol/L sodium acetate
asmobile phase,5 compositions (estriol,estradilo,estrone,
prgesterone,testosterone) canbe successfully separated. Between 1.0μ
g/mL and 3μg/mL,the linear relationshipof standard curve was
satisfied for each composition (Relativc Coeffieient >
0.999).Thedetection limit respectively were: estriol
0.009mg/kg;estradiol 0.038mg/kg; estrone0.044mg/kg; testosterone 0.
02mg/kg; progesterone 0.071 mg/kg.The recovery rate forevery
composition in different content level was more than 80%,The
coeffieient ofvariation obtained for every composition was less than
l5%。 This method is simple,accurate and suitable for routine
