In our experiment Kunming mouse peritoneal cavity macrophages had been cultured fora few days and the cells were divided into four groups,placed at the temperature of 37C 139C,41 C and 43C for 2 hours respectively.The 37oC group was used for control and other,three groups,for experiment.The fluorescence intensity of DNA molecule in the cells ofeach group was measured with Adherent Cell Analysis and Sorting interactive Laser Cytometry 570(ACAS 570) after staining with the DNA-specific dye 4,6-diamidino-2-phenylindole(DAPI).The integrated and average fluorescence value as well as the relationship betweenthe average fluorescence value and cell area were taken as the main indexes of the measurement.The results showed that the fluorescence intensity of 39C-treated cells was lowercmpared to that of control(37C) ones but it raised in 41 oC-treated cells and reached thehighest in 43oC-treated cells.Thus, it was considered that the changes of f.luorescence intensity of DNA molecule in cultured cells may represent some undergoing processes of the cellscaused by the heat-treatment(39 C ̄43 C )
