摘要 用扁桃腺细胞诱生白细胞介素-2(IL2),较适宜的条件是细胞数5×106/ml,PHA100μg/ml,加白细胞干扰素200IU/ml起动,37℃5%CO2培养36~48小时,pH不低于7.0,能较相对稳定地获得平均活性>700~1,200IU/ml的IL2,最高效价可达2,200IU/ml。以诱生的粗IL2(2,000IU/ml),用硫酸铵80%盐析及DEAN.Sspharose离子交换层析二步法部分纯化,比活性即可达即达>3.0×106IU/mg。回收率≈60%。本法简易、快捷,用作提纯IL2是较好的方法。 This study was on the induction and purification of human IL-2 produced by tonsil lymphocytes. We found that the optimal cnditions for IL-2 induction were: Cells from tonsil in solution pH≥7.0 and at the concentration of 5×106/ml, adding PHA 100μg/ml, initiation IFN 200IU/ml, incubating the cells at 37℃ for 36-48 hours under 5%CO2 atmosphere. The average titer of Such induced IL-2 got over 700-1, 200IU/ml and the highest titer reached 2, 200IU/ml. The coarse IL-2 (2, 000IU/ml) was partly purified with (NH4)2SO4 precipitation and DEAE-Sepharose chromatography, the specific activity of purified IL-2 was over 3.0× 105IU/mg, receiving rate 60%. We regarded this method as a simply, rapid and good one for purification of nature IL-2.
