小鼠胎肝基质细胞高活性造血刺激因子的纯化及性能研究

摘要 对我室克隆的小鼠胎肝基质细胞林(MFLSC株)的无血清培养上清中的造血活性物质进行了初步纯化。通过离子交换层析和凝胶过滤层析两步分离,从中得到一高活性的造血刺激因子,称为基质细胞造血刺激因子-1(SHF-1)。经凝胶过滤及电泳结果估测,此因子是分子量约为70kD的单体蛋白,耐碱,较耐热。在体外它对小鼠骨髓粒系、红系祖细胞、多向祖细胞及多能造血干细胞均有刺激作用。 A hematopoietic growth factor was purified from the serum-free conditioned medium of a murine fetal liver stromal cell strain ( MFLSC) by using techniques of ion-exchange and gel filtration chromatography. It was named stromal cell hematopoietic growth factor-1 (SHF-1). Estimated by gel filtration and electrophoresis, the molecular weight of SHF-1 was approximately 70kD and SDS-PAGE showed it was a monomeric protein. In vitro it not only stimulated the proliferation and differentiation of murine marrow granulocyte-macrophage, erythroid and multipotential progenitor cells, but also triggered resting spleen colony forming unit(CFU-S) into cell cycle.
