摘要
本文研究了培养不同时间的骨髓基质细胞对粒系祖细胞体外培养生长的影响,并分析其细胞成分及培养上清液的作用。结果表明:在含上清的基质细胞贴壁层上种植骨髓细胞,除第14天外,均可见对CFU-GM的形成有刺激作用。而单纯无上清贴壁细胞层,或单纯基质贴壁细胞培养上清液,均无此刺激作用。说明基质细胞对粒系造血祖细胞生长的调控必须在一个结构功能完整的体系中才能发挥作用。在无外源性CSF作用的情况下,基质细胞仍能使少量CFU-GM生长,也说明基质细胞有刺激粒细胞生长的作用。14天以前,无琼脂间隔层的粒系祖细胞数较高,而21天后,有琼脂间隔层的粒系祖细胞数较高。说明造血基质细胞对粒系细胞生长的调控受着多方面因素的影响,体外培养时间不同有不同表现。基质细胞一方面通过与粒系造血祖细胞相互作用,近距离调节,另方面通过分泌粒系刺激因子和抑制因子,从正、负两方面调节粒系造血,以维持粒系造血的动态平衡。
The effects of bone marrow stromal cells on growth of the granulopoietic progenitors were investigated in vitro. The actions of the stromal cells in the adherent layer and the supernatant of the culture were analysed. It was found that when the bone marrow cells were implanted onto the stromal cell adherent layer containing the supernatant the stimulation to CFU-GM formation was observed except on the 14th day of culture, while low stimulating effect on the CFU-GM formation existed when either the adherent stromal cells or the supernatant alone were added. These facts suggested that the modulation of bone marrow stromal cells on the growth of granulopoietic progenitors could be achieved mainly through a system with integrated structure and function. Under the condition of no exogenous CSF, the stromal cells could still stimulate the formation of a small amount of CFU-GM, which implies that the stromal cells possess the capability to stimulate the growth of granulocyte progenitors. The number of granulopoietic progenitors without the separation of a thin agar layer was higher before the 14th day of culture while that with an agar layer separation was higher after the 21st day, which indicated that the modulation of bone marrow stromal cells on the proliferation of granulopoietic progenitors was influenced by more than one factor. Different results were obtained when the time of culture varied. The stromal cells practised their modulation through their stimulation to the CFU-GM formation and the interactions with the granulopoietic progenitors on the one hand and through the secretion of granulopoietic stimulating or inhibiting factors on the other. Thus the proliferation of granulopoietic progenitors is adjusted both positively and negatively to maintain a dynamic balance of granulopoiesis.
