摘要
本文以辣根过氧化物酶(PO)作抗原与相应抗体(抗-PO)混合,加入血清,对血清补体抑制免疫复合物沉淀作用(IIPC)进行了研究。结果表明,在抗原抗体最适比例时形成免疫复合物最多;原血清及1∶2稀释血清IIPC最强,OD值为0.57和0.54,血清的进一步稀释对IIPC有一定影响;孵育时间以30min最宜,OD值为0.56;离心时间在25~45min时OD值最大,为0.50~0.56;在最适条件下,正常人混合血清OD值为0.49;56℃30min灭活血清为0.14;50℃15min处理血清为0.38;ED-TA处理血清为0.09,EGTA处理血清为0.21;对20例健康人血清IIPC测定结果为0.39~0.62,平均0.48±0.07。表明正常血清补体对免疫复合物的沉淀有明显抑制作用,各种因素处理血清对IIPC均有一定影响,而以56℃30min和EDTA处理血清最明显。文中还对该法的影响因素进行了讨论。
In this paper, a method for measuring inhibition of immune precipitation by complement(IIPC) using peroxidase (PO) as a antigen was studies. The results showed that immune complex of PO and anti-PO rabbit IgG at equivalence could form a great quantity of precipitation. OD values were 0. 57and 0. 54 in non and diluted serum(1: 2 ), 0. 56 in 30 minutes incubation time, and 0. 50~0. 56 in 25~45 minutes centrifugation time. OD value of normal individuals mixture seruml the serum of heating to 56℃ for 30 minutes, 50℃ 15 minutes, and treated by ethylenediamine tetracetate (EDTA), ethyleneglycol tetracetate(EGTA) were 0. 49, 0. 14, 0. 38, 0. 09, 0. 21 respectively. IIPC of 20 normal persons was 0. 48 ± 0. 07 (0. 39 ~ 0. 62). These results indicate that normal serum markedly inhibites precipitation of immune complex and many agents may block IIPC of serum, especially 56℃ for 30 minutes and EDTA. The various factors of inhibiting IIPC are discussed in the paper.
