摘要
我们检测了6例活动期、6例缓解期SLE患者和10例正常人血清和外周血单个核细胞(PBMC)自发及经天然DNA(nDNA)、rIL—2、rIL—6和rIFN—γ诱导后分泌IgG类抗双链DNA(dsDNA)特异性抗体水平,观察到活动期患者血清和自发分泌量显著高于缓解期,后者又显著高于正常人,且体外细胞分泌平与血清中的含量密切相关(r=0.9882)。nDNA能特异地激发SLE患者的FBMC分泌抗dsDNA抗体,但对正常人无作用。rIL—2和rIFN—γ皆能增强SLE患者PBMC产生抗dsDNA抗体,表明即使缓解期病人,血中仍有表达IL—2受体的激活的B细胞存在。nDNA和rIL—2联合应用时,对活动期患者PBMC分泌抗dsDNA抗体有协同作用。本文未见外源性rIL—6对抗dsDNA抗体分泌的影响。
The amount of specific antidouble stranded DNA ( anti-dsDNA) antibodies of IgG class in sera and culture supernatant of peripherial blood mononuclear cells (PBMC) spontaneous secretion and stimulated by native DNA ( nDNA) and cytokines ( rIL-2, rIL

is shown that the level in sera and spontaneous culturing supernatant of PBMC from active SLE pttien,.; is significantly higher than that of inactive subjects, the latter is markedly elevated while comparing with that of normal donors.The level of anti-dsDNA secretion in vitro show more closely correlation with that of in sera (r = 0.9882) .nDNA can specifically stimulate PBMC of SLE patients to secrete anti- dsDNA antibody but no effects to normal's lymphocyte. - dsDNA antibody production by PBMC from active and inactive SLE patients. It indicates that there are a lot of activated B lymphocytes expressed IL-2 receptor in inactive patients' blood as well as in that of active patients. We also find that nDNA antigen can cooperate with rIL-2 to induce anti-dsDNA antibody secretion by active SLE PBMC .We don't find the obvious effect of exogenous rIL-6 on anti-dsDN antibody secretion.
