快速微量Dot—IGSS检测日本血吸虫病人血清中抗体的研究 被引量:4

摘要 本文用日本血吸虫可溶性虫卵抗原点于微孔滤膜上,以快速微量斑点免疫金银染色法(Rapid microvolume Dot-IGSS,Rm Dot-IGSS)检测血吸虫病患者血清中相应抗体,并与Dot-IGSS相比较,检测血吸虫病人血清69份,两法阳性率均为100%。检测正常人血清50份,两法阴性率均为100%。检测华支睾吸虫病人血清10份,两法均未出现交叉反应。检测肺吸虫病人血清10份,Rm Dot-IGSS未出现交叉反应,Dot-IGSS有1例出现交叉反应。 Having achieved good results in detecting serum antibody from patients with clonorchiasis and schistosomiasis by immunogold-silver staining (IGSS) technique, further study was carried out on diagnosis of schistosomiasis japonica by rapid microvolume Dot-IGSS or Rm Dot-IGSS. Serum samples from 69 schistosomiasis patients, 50 normal individuals, 10 paragonimiasis westermani patients and another 10 from clonorchiasis sinensis patients were tested by IGSS us- ing soluble egg antigen of Schistosoma japonicum (protein content 2 mg/ml). The microvolume of antigen was added into the samll square of filter paper,adopting the method of Dot-IGSS of Wu Zongxing et al. Simutaneously,Dot-IGSS was made for comparison. Sixty-nine serum samples of schistosomiasis patients were all positive for Rm Dot-IGSS and Dot-IGSS. Fifty normal serum samples were all negative. Ten serum samples from clonorchiasis patients were all negative by both methods. One serum sample from paragonimiasis had cross-re- action for Dot-IGSS but all negative for Rm Dot-IGSS. This method (Rm Dot-IGSS)only needs 1/5-1/15 amount of serum or reagent as compared with that of Dot-IGSS and 30 minutes for handling the whole test.
