摘要
本文应用 Western 印渍和原位免疫荧光定量技术,检测了两株经 EBV-DNABamHI-W(YH)片段转染后转化而建立的细胞株,CH-1和 CH-2所表达的 EBNA 亚型.结果发现,两株细胞均有一系列由27~38KD 小分子量的 EBNA5表达,同时可见 CH-1细胞有低水平的 EBNA2表达.对比起无 EBNA2表达的 CH-2细胞,CH-1细胞具有较低的接触抑制能力和附壁性,并且细胞蛋白质的代谢也有较明显的改变,提示 EBNA2和 EBNA5均与细胞转化有关。
By means of Western blotting and immunofluorence quantity in situ,we have detected Epstein --Barr virus nuclear antigens(EBNAs)expressed in two transformed cell lines,CH--1 and CH--2, which were transformed by transfected with EBV--DNA BamHI--W(YH) fragment.The result showed that both cell lines expressed EBNA5 in a series of low molecular weight ranging from 27,~ 38 KD.In addition,the CH--1 cell line,which exprssed EBNA 2 in low level,was of lower contact inhibition,higher anchor independence and greater change in cellular protein metabolism than the CH--2 cell line without EBNA2 exprssion.These results suggest that EBNA2 and EBNA5 are asso- ciated with cell transformation.
