摘要
实验研究海洋新药“916”对动脉粥样硬化的预防作用。结果表明,“916”能明显降低高脂血症鹌鹑血清总胆固醇(TC),低密度脂蛋白胆固醇(LDL—C),升高高密度脂蛋白胆固醇(HDL—C)及其亚组分Ⅲ(HDL_2—C),减少过氧化脂质(LPO)代谢产物丙二醛(MDA)生成,降低胆固醇,特别是胆固醇在动脉壁中的沉积,抑制动脉壁LPO生成,从而减少动脉粥样斑块的生成,能延缓和阻止了动脉粥样硬化的发生和发展。。
'916' is a new type of low molecular weight heparinoid based on Chitosan.In this paper the preventive effects of '916' on experimental atherosclerosis were studied.The results show that '916' can significantly inhibit the elevation of serum TC and (VLDL+LDL)-C and increace HDL-C/TC,HDL3-C and inhibit the enhanced serum MDA.The TC and CE contents in arterial wall in '916' treated groups were dramatically lower than those in hypercholesterolemic group at the end of the experiment,together with the decrease in the ratio of CE to TC.The contents of MDA in arterial wall in the treated group was remarkably lower than the hypercholesterolemic group,indicating that '916' could inhibit the deposit of cholesterol,especially cholesterol ester in arterial wall and decrease the production of lipid peroxides.The lesion in the hypercholesterolmic group was more severe with incidence of antherogenic plaques 91.67% and those in the '916' treated group much milder as compared with the hypercholesterolemic group.Histological examination showed that there appeared proliferation of smooth muscle cells with large amounts of foam cells in intima,while fewer proliferated smooth muscle cells and foam cells were observed in the '916' treated group.In conclusion,' 916' can prevent the development of atherosclerosis through modulating lipoproteins and inhibiting the production of lipid peroxides.
