间羟舒喘灵诱发豚鼠肺组织β受体下调的动态观察

摘要 本实验采用~3H—DHA放射配基结合法,动态观察了β—受体激动剂间羟舒喘灵对哮喘豚鼠肺组织β—肾上腺素能受体(βAR)的作用。结果表明:治疗早期豚鼠肺组织βAR无明显变化,随着时间的延长(5天以上),βAR显著下调。以上结果提示:支气管哮喘βAR功能低下并非疾病的原发因素,而是继发治疗的结果。βAR下调可能是患者对β—受体激动剂失敏的重要原因。 Using radioligand binding assay, We investigated the effect of terbutaline on beta-adrenoceptors density of Guinea pig lung tissue in experimental asthma induced by ovalbumen, The results showed that: 1)there was no significant difference between the normal control and asthmatic control group, 2) short time inhalation of terbutaline did' nt reduce the density of beta-adrenoceptors in lung tissue, 3)the density of beta-adrenoceptors was reduced markedly after long term treatment, 4) terbutaling did'nt sig nificantly change the Kd value. These findings sugg est that the decreased betaadrenoceptors density in asthmatics be caused by therapy with agonist rather than an inherent feature of the disease, and may reflect tachyphylaxis from the effect of long term inhalation of terbutalin therapy.
