摘要
用分泌抗-HBs Ag单克隆抗体的杂交瘤细胞的8-氮杂鸟嘌呤(8-AG)耐受株作为亲本细胞,与辣根过氧化物酶(HRP)免疫的BALB/c小鼠脾细胞进行第2次融合,从382个克隆上清中筛选出一株分泌抗-HBs Ag-抗-HRP双特异抗体的阳性孔。经多次克隆纯化建立3株杂交-杂交瘤,命名为Bs33、Bs38和Bs39,其抗体均属Ig G_1型。细胞染色体平均为113条。细胞反复冻存、复苏、传代仍稳定分泌双特异抗体。
8-azaguanine (8-Ag) resistant hybridorna mutants keeping the ability of secreting anti-HBsAg McAb were used to fuse with the spleen cells of BALB/C mice which had ben immunized with horseradish peroxidase (HRP). One positive clone secreting anti-HBsAg anti-HRP bispeeifie MeAb was obtained screening from 328 clones. 3 hybrid-hybridomas, named BS33, BS38 and BS39, were obtained by consecutive clone purification. All the bispecific McAbs were IgG, subclass and the mean of chromosornes of hybrid-hybridorna was 113. After repeatedly frozen, thawed and passased, the three hybrid-hybridomas were proved to be capable of secreting bispecific McAb stably.
