基因重组α型干扰素对人白血病细胞生长增殖的抑制作用

摘要 白血病细胞与干扰素共同培养后,~3H-TdR和~3H-亮氨酸掺入率明显减低,表明干扰素抑制白血病细胞的DNA和蛋白合成。对白血病细胞生长增殖动态及细胞周期研究表明,干扰素可以延缓白血病细胞的生长增殖,阻滞白血病细胞从G1期向S期的转化过程。 Growth and proliferation inhibition action in recombinant interferon-α (rIFN-α) was studied on two human B iymphoma leukemic cell lines (Raji, Daudi). After co-culturing leukemic cells with rIFN-α,the rate of incorporation of ~3H-TdR and ~3H-leucine into the cells was significantly decreased in both leukemic cell lines indicating an inhibition of DNA and protein synthesis in the cells. Further study of cell growth dynamics and cell cycle analysis showed that rIFN-α could inhibite cell proliferation,and block the process of G1 to S phase transition. Thus,rIFN-α may mediate its anti- leukemia activity via direct inhibitory effect on growth and proliferation of leukemic cells.
