高效液相色谱法同时测定人尿中假尿核苷和肌酐

摘要 采用反相高效液相色谱同时测定人尿中假尿核苷(ψ)和肌酐(Cr)。尿样经过滤、稀释后可直接进样,具有简便、快速和准确的特点。并探讨了ψ/Cr 值与肺癌的相关性.利用本法测定了69例正常成人,平均值为23.1±4.4μmol/mmol,22例肺部感染患者为26.2±4.2μmol/mmol,43例肺癌患者为32.8±7.9μmol/mmol。肺癌患者比肺部感染者及正常者有显著增高(P<0.001),同时讨论了ψ/Cr 值作为一种新的肺肿瘤标志物的临床意义。 A simple procedure for the simultaneous determination of pseudouridine and creatinine in urine using reversed-phase high performance liquld chromatography with ultraviolet detection is described.It consists of simply diluting the filtered urine with mobile phase (1∶20)followed by dire- ct chromatograhic injection.This method has been applied to the analysis of urine samples from normal donors and patients with lung cancer and pul- monary infections.The mean urinary pseudouridiue concentration was signi- ficantly higher in the patients with lung cancer (32.75±7.92((?)+s)μmol/ mmol) than that in patients with pulmonary infectious diseases (26.25±4.17 μmol/mmol) or in healthy controls (23.06±4.41μmol/mmol).These findings indicate that urinary pseudouridine may be a useful biochemical marker in patients with lung cancer.
