36只Wistar雌性成年大鼠随机分为单用内毒素、单用头孢唑肟及头孢唑肟+内毒素和对照组。分别于腹腔内一次性注入后1h处死动物,结果示各处死组血BUN,Scr和肾酶组织化学与对照组比较差异不显著。光镜下可见其病理改变,尤以头孢唑肟+内毒素组为重,并有血和肾组织匀浆中TXB_2/6—keto—PGF_(1α)比值增大,提示头孢唑肟与内毒素两因素叠加时可增加肾毒性,但其改变早期不能以酶组化和BUN,Scr所显示,确以TXB_2/6—keto—PGF_(1α)的改变敏感而可靠。
36 adult female Wistar rats were randomly divided into endotoxin(E), ceftizoxime(C); endotoxin with ceftizoxime (E+C) and control groups. All rats were killed 1 hour after abdominal cavity injection. Statistic analysis showed no significant difference in serum creatinine and BUN among these groups (P>0.1,>0.2).There were little changes in enzyme activity of tubular cell mitochondria in experimental groups at early stage, but swelling of tubular cells, deduction of matrix and swollen of mitochondria were prominent. Meanwhile TXB 2/6-keto-PGF1αincreased in serum and renopareachymal hemogenate. These changes were most prominent in E+C groups TXB2 in serum were 7.12 times higher than control group while 6-keto-PGF1α were 41 per cent lower than control (P<0.01,<0.05).Concomitant occurrence of ceftizoxime and endotoxin might destroy the 'integrity of vascular function' and balance of TXB2/6-keto-PGF1α, thus result spasm of vessels. Therefore above ultrastructural changes appeared as the result of interstitial ischemia and hypoxia due to the decrease of renal blood flow and glomerular filtration rate.
