小鼠Kupffer细胞自发产生肿瘤坏死因子及BCG对它的激活作用 被引量:4

摘要 由正常昆明(KM)小鼠肝脏分离的非实质性细胞(NPC)和 Kupffer 细胞(KC)以及脾巨噬细胞(SMφ)在体外培养上清中能检测到对 YAC-1细胞有细胞毒作用的肿瘤坏死因子(TNF),但对L_(929)细胞无细胞毒作用。表明肝和脾的巨噬细胞能自发产生一定水平的 TNF,但它的作用有选择性。用 BCG 和 LPS 给小鼠腹腔注射在原位激活后,KC 和 SMφ所诱生的 TNF 对 YAC—1和 L_(929)细胞都有细胞毒作用。这些结果提示 KC 在肝脏内防御转移的肿瘤细胞生长起重要作用。 The cytotoxicity of the TNF against YAC—1 cells was detected in the cell culture su- pernatants of liver nonparenchymal cells(NPC),Kupffer cells(KC)and spleen macrophages isolated from kunming(KM)mice.But this substances had no cytotoxic activity against L_(929)cells.These re- sults suggest that the macrophages of liver and spleen of mice can spontaneously produce a given levels of TNF.After in situ activation by the systemic administration of BCG and LPS,the TNF induced by KC and spleen macrophages were all able to cytotoxic against YAC-1 and L_(929)cells.These studies indi- cated that the liver fixed macrophages may play important role in host defense against tumor cells that metastasize to the liver.
