人皮肤细胞培养对同种异体创面移植的临床研究

摘要 本文是在人表皮细胞培养研究和临床应用的基础上,进行真皮成纤维细胞培养,并将其与表皮细胞分期移植于烧伤创面,收到满意效果。 Since 1984, we have created a biological skin equivalent consisting of cultured ker- atinocytes,and the skin equivalents were applied to 11 patients with burns.Since 1990,another biologi- cal skin equivalent consisting of cultured fabrolasts have been created,and we grafted respectively the skin equivalent of cultured fabroblasts and the skin equivalent of cultured keratinocytes onto same sur- face of skin de-fect in 6 patients with burns.The results were satisfactory. The skin biopsy samples lcm in diameter were obtained at routine circumcision,and epidermis and dermis were separated with trypsin,the keratinocytes and fabroblasts were cultured respectively in a rat. tail collagen with RPUI 1640.The keretinocytes were cultured for 4-5 days,and the fabroblasts were for 2 weeks.At early stage.We grafted simply the skin equivalents of cultured keratinocytes onto the surface of skin defects in 11 patients with burns.Since 1990,we have used a new method in 6 patib- nts.At first,the skin equivalents consisting of cultured fabroblasts were grfted onto a surface of skin de-fects with burns,one week late another skin equivalents consisting of cultured keratinocytes were graft-ed onto same areas.The skin equivalent of cultured keratinocyres and a dermal component is without the constituents that elicit a pronouoced immune response.Compared the grafts by using two different methos under the light microscope,although the grafts by using two different methods can survive easy,we found,the grafts by using the method that a skin equivalent of cultured fabroblasts and a skin equivalent of cultured fabrodlasts and a skin equivalent of cultured keratinocyte were graft- ed respec-tively onto same area of skin defact is more substantial and compact than by using the method in which the skin equivalent of cultured keratinocytes was grafted simply.
