人体乳腺癌细胞系BGaP-37和MGF-7的细胞遗传学研究

摘要 本文应用普通染色体G一显带技术和高分辨染色体显带技术对两个人体乳癌细胞系进厅了详尽的细胞遗传学研究。结果显示两个细胞系均具有较复,桀的染色体结构和数目_}辛常。Bcap一3 7细胞系染色体众数为62~63,具有23个可识别其结构的标记染色体。MCF一7细胞系染色体众效为55—56,具有l9个可识别其结构的标记染色体,进一步分析认为,第1、3、5、7、11、13和17号染色体的数目和结构改变,可能与乳腺癌的发生和演进有一定联系;染色体断点1p^(11)(1q^(11))、1p^(13)、3p^(21)、3q^(11)、5q^(11)、7p^(13)、7q^(22)及导致11p^-、17p^-6q^-、13q^-的断裂点可能通过癌基因的激活和抗癌基因的丢失而起一定怍用。 Cytogenet c stud es of human breast,cancer ce lines( Beap—37, MCF—7)were carried out with G—banding and high resolution chromosome banding thchniques The results demonstrated extensive aneuploidy with both numerical and structural abnormalities in both cell lines It was found that Bcap—37 cell line had 62—63 chromosomes with 23 identified markers in most examined cells The modal chromosome numbers in MCF-7 cell line Was 55—56,and 19 identified markers were found in MCF—7 cell line The results suggested that the numerical and structural changes of chromosome 1,3,5,7,11,13 and 17 might be associated with breast cancer, and that the breakpoints 1p11(1q11),1p13,3p21,3q11,5p11,7q13,7q22 and the Points resulting in 11P—,17P—,6q—,13q—may be reiated to the cxprossion of activated oncogenes and loss of antioncogenes
