摘要
经给予大白鼠不同的针刺时间(1、2、4、6、8、10、12hr)后灌流杀死取材,采用定量电镜方法观察记数了三叉神经尾侧脊束核胶状质亚核内大单壁,尤其是双壁有衣小泡(DWCVs)的形态,数目及不同针刺时间内数量的变化规律。结果发现大单壁有衣小泡的数量与针刺时间无明显正比关系,而DWCVs不但形态多样化,其数目也随针麻时间均不同而改变。本文依据Eckenhoff的分类方法将DWCVs归纳为以下5种类型:(1)尚未与相邻两终末质膜脱离的孤立的DWCVs;(2)游离于终末内孤立的DWCVs;(3)与终末质膜相连的融合的DWCV簇;(4)游离于终末内融合的DWCV簇;(5)树突棘凸入另一轴或树突内并与之共同形成的不典型的DWCV簇。本研究还发现,在针刺麻醉中DWCVs在1~8小时内的形成与时间成正比,即1小时始增多,4小时显著增多,8小时达高峰,此后开始下降,10小时已回到4小时的水平,直至12小时才恢复到针麻前的状态。对照组仅见少数孤立的DWCVs,其形成基本不随时间而变化。
The experimental rats were killed by perfusing the brain through ascending aorta with fixative solution containing 2% polyformaldehyde and 0.5% tannic acid following acupuncturing the points of Ren Zhong and Si Bai for Various time, and the specimens were obtained from the substantia gelitinosa of the caudal trigeminal nucleus. Using quantitative electronmicroscopic method, we studied the formation, morphological features and variations in number about coated vesicles. The results show that the large single-walled coated vesicles are not increased obviously in number with time, while the double-walIed coated vesicles (DWCVs) are not only formed in quantity but their shape also changed correspondingly. Acording to Eckenhoff, all of the DWCVs can be divided into five types. They are: (1) solitary DWCVs, attached to the surface membranes; (2) solit ry DWCVs, free in cytoplasm; (8) DWCVs in fused elusters, attached to the surface membranes; (4) DWCVs in fused cluster, free in the cytoplasm; (5) the un-typical fused cluster, which are presumed that a dendritic spine protruede into other terminals and formed several solitary DWCVs together with the terminal membrane at one side. The results also indicated that different time of acupuncture anaesthesia, represented by abscissa, showed various effects on the DWCV forming number, ordinate (Fig. 1). In the Fig. 1, the number of DWCVs tended to rise in a linear function within 8 hours, i. e. the DWCVs occurred in terminal increasingly in the first hour, increased markedly from the 4th hour, and reached the maximum at the 8th hour; then begun to decrease in number, at the 10th hour returned to the level of the 4th hour, and until the 12th hour got back to the original state.
