摘要
以黑曲霉、米曲霉、木霉、阿舒假囊酵母等几种工业生产用重要真菌为实验对象。通过研究其菌龄、酶的选择及配比、酶解时间等对原生质体制备的影响,以及再生频率的大小和再生过程的显微摄影观察,初步摸索到酶法制备上述丝状真菌原生质体及其原生质体再生成菌体的条件。从而为在原生质体水平上对这些菌株进行遗传操作,尤其是外源基因导入这些菌株对其进行遗传改良提供了实验基础。
This paper reports the optimal conditions of protoplast formation and regeneration of Aspergillus niger 3.396, Asp. niger 3324,Asp.niger C_1—I_(13),Asp.oryzae Xi—3, Trichoderma sp. and Eremothecium ashbyii We studied factors affecting protoplast formation, such as the optimal age of mycelia, the selection and mixing proporation of different kinds of enzymes, and the time of enzymolysis; caculated the frequencies of regeneration and observed the process of regeneration with microphotography, The regeneration frequencies were 0.12%—3.48%.
