转化前后K-562细胞胰岛素受体酪氨酸蛋白激酶活性及内源性底物的研究

摘要 对丁酸钠转化前后的人红白血病细胞林K-562的表面胰岛素受体数量、胰岛素受体酪氨酸蛋白激酶活性及细胞内源性底物进行了研究。结果表明:丁酸钠转化后的K-562细胞酪氨酸蛋白激酶活性降低,胰岛素受体数量减少,K-562细胞酪氨酸蛋白激酶的内源性底物,分子量为67KD—底物蛋白在转化后消失。说明该底物及胰岛素受体酪氨酸蛋白激酶在胰岛素调控的K-562细胞的增殖中起重要作用。 In this paper, the quantity of insulin receptors and the receptor tyrosine kinase activity and its endogenous substrates were comparatively studied in K-562 cells before and after transformation. It was shown that following transformation, the quantity of insulin receptor and the insulin-stimulated tyrosine kinase activity were reduced distinctly, and the endogenous substrate, a 67KD protein for the receptor tyrosine kinase disappeared. These results indicate that the insulin receptor tyrosire (?)inale and its endogenous substrate may play an important role in the insulin-controlled porliferation of K-562 cell.
