摘要
研究了培养基、血清浓度和培养温度对山羊半胚体外发育的影响。结果表明:与杜氏磷酸缓冲液和TCM—199相比,在改良Whitten氏液中进行体外培养可显著提高半胚体外发育率(P<0.05);用改良Whitten氏液进行体外培养,含10%和15%犊牛血清组的半胚发育率显著高于5%血清组(P<0.05),极显著地高于无犊牛血清组(P<0.01);在38~39℃的条件下培养半胚的效果显著优于36℃(P<0.05),但在37~39℃的范围内,温度对半胚体外发育率无显著影响(P>0.05)。
The effect of medium types, fetal calf serum (PCS) concentration and temperature for culture on in vitro development of goat demi-embryos was studied at the present study. The results are as follows: modified Whitten's medium suported the development of the demi-embryos significantly better than Dulbecco's phosphate buffered saline and TCM-199 (P<0.05) did; the developmental rate of demi-embryos was significantly highter in modified Whitten's medium with 15% and 10% PCS than with 5% (P<0.05) and 0% PCS (P<0.01); higher survival rate of demi-embryos in vitro could be obtained when culture at 38 in comparison with at 36 (P<0.05), but no significantly different results were found when culture at between 37-39 (P>0.05).
