铝氟及莫混合液大鼠小肠原位灌流对酶活性的影响

Effects of Aluminum, Fluorine and Their Mixtures on the Enzyme Activities of Jejunal Mucus Membrane in the Perfusion of Small Intestine of Rats in Viro

摘要 本实验用 Al 32mmol/L,Al 170mmol/L,F 100mmol/L,Al32mmol/L+F100mmol/L,Al 170mmol/L+F100mmol/L五种溶液,分别原位灌流大鼠小肠40分钟,取空肠上段作组织学和15种酶的组织化学观察。结果显示:氟对脱氢酶(SDH,ICDH,NADHD,MDH,GDH,GPDH,LDH),氧化酶(CCO)和部分水解酶(ANAE,G 6 Pase,5′-N,ATPase,AlP,AcP)呈不同程度的抑制作用。铝则主要抑制脱氢酶及氧化酶。一定的铝氟混合后,酶活性较相应单纯铝及氟组均有升高。 Buffer (control), F as NaF(100mmol/L), Al as AlCl_3, (32 and 170mmol/L) and the two mixtures of Al and F(Al:F=0.3:0.7 and 0.7:0.3) in Tris-bufferedmedia of pH7.0 were respectively perfusedthrough the small intestine of rats over aperiod of 40 min. The jejunum was takenout from each animal immediately. Someof its parts were for HE staining; and theothers for enzyme histochemical staining. Fifteen enzymes were observed. The resultsshowed that (1) Fluorine chiefly inhibitedthe metal-enzymes and metal-activated en-zymes; aluminum mainly depressed the ac-tivities of enzymes in relation to the cellenergy metabolism, such as hydrogenasesand oxidase. (2) The decreased activitiesof enzymes induced by A1 or F increasedsignificantly after mixing with each other,indicating that the simultaneous existenceof A1 and F decreased their ionic concentra-tions, so that strong antagonistic effects onthe inhibition of enzymes were noticed sinceF or Al inhibited enzyme activities by theirions. (8) A high dose of Al intensively in-hibited the enzymes related to the cell en-ergy metabolism, which explained why lessquantity of A1 was absorbed in animals withhigh dose of Al than that of low dose A1seen in the absorption experiment. (4) Theenzyme activities in the animals given highdose of A1 mixed with F apparently wereelevated in comparison with that in the ani-mals given a pure high dose of A1 whichwas the result of antagonistic effects of F onthe inhibition of enzyme activities by A1,therefore the quantity of the absorbed A1was high.
