萘乙酸诱导蚕豆种子愈伤组织的机理初探

摘要 用1.0~5.0ppm的萘乙酸(NAA),可诱导蚕豆发芽种子全部产生肿胀状愈伤组织,并且抑制芽的伸长。转录抑制剂放线菌素D20ppm处理蚕豆种子时,不影响发芽率,但抑制芽的伸长。当用放线菌素D与NAA复合处理蚕豆种子时,发芽率也不受影响,但芽的伸长明显受到抑制,此外还有55%左右的发芽种子胚轴基部形成肿胀状愈伤组织。用5.0ppm的环己亚胺处理蚕豆种子时,所有种子均不发芽。结合对酯酶同工酶谱的分析,作者认为,NAA诱导蚕豆种子产生愈伤组织是在翻译水平上的调控,而不是在转录水平上。 By using of RNA synthesis inhibitor actinomycin D. (ACTD) and protein synthesis inhibitor cycloheximide(CHM), the effect of NAA on seed callus induction of broad bean was studied. It showes that the seed can germinate from the medium which contained ACTD(20 ppm) but the growth of germinative seed was inhibited. And NAA(1.0—5.0 ppm) was observed to induce callus from all germinative seed and to inhibit the growth of bud also. The ability of germination of seed in the medium contained CHM(5.0 ppm) has not been observed, In the medium which contained varying concentration of ACTD in combination with NAA, the growth of bud of germinative seed was inhibited obviously and callus still be induced from the germinarive seed. The esterase isozyme pattern showed that the addition of ACTD inhibited the appearance of a band(Rf= 0.573). The results mentioned above showed that there were long lived mRNA in the seed and it suggested to us that the effect of NAA in seed callus induction was probably acted on translation level.
