单克隆抗体对甲_1型流感病毒的抗原分析和快速诊断的研究

摘要 用宁夏地区分离的甲,型流感毒株宁防86—27(H_1N_1)制备了抗甲_1型流感病毒血凝素的McAb39株,其血抑滴度在1:2000—1:256000,Ig亚类为IgG_1、IgG2a、IgG2b。用自制的McAb对宁夏历年分离的甲_1型毒株的抗原分析表明:抗原漂移存在,且以1981和1987年的毒株抗原漂移明显。用我们的McAb对甲_1型毒株的间接免疫荧光试验,证实此法特异性好,敏感性高,确有早期快速诊断的意义。 Thirty-Nine Monoclonal antibodies (McAbs) specific for the hemaglutinin of A/Ningxia/27/86 (H1N1) were developed. The Hemaglutination inhibition titer was 1: 2000-1: 256000. The neuraminidase inhibition titer of these McAbs was less 1: 10. None of the McAbs reacted with influenza virus H3N2, type B and C. 23 McAbs were of subclass IgG1,9 of IgG2a and 7 of IgG2b, respectively. Antigenic analysis of H1N1 influenza virus isolated in 1977, 1981, 1982, 1984, 1986 and 1987 in Ningxia was carried out by using 39 McAbs. The results showed that there were two antigenic variations, one in the 1981 isolates, and the other in the 1987 isolates. All McAbs failed to react with either the 1981 or the 1987 isolates. A, comparison of the sensitivity of the immuno-fluorescence (IF) test using a McAb with different doses of A/Ningxia/27/86 was done. With a virus dose of 10^(-1), fluorescence was seen as early as 4 hours after infection, while CPE occurred at 8 hours. With a virus dose of 10^(-3), fluorescence could be seen at 8 hours, Whereas CPE occurred at 24 hours.
