DNA多聚酶连锁反应(PCR)研究和应用现状

摘要 自SAIKI等1985年首次将DNA多聚酶连锁反应(The polymerase chain reaction,PCR)用于镰状红细胞贫血诊断以来.短短五年,这一DNA扩增技术已日趋完善,并日益广泛应用于分子生物学、遗传学、病毒学、免疫学、法医学研究和临床诊断各领域。它不必经常规程序分离DNA和分子克隆,便可在若干小时内将待检DNA扩增约一百万倍,既可在琼脂糖凝胶电泳时以溴乙锭(Ethidium bromide)染色及紫外线照射直接观察,又可用于核酸杂交或序列分析;且极其敏感和迅速,所以被誉为“革命性的变革”。
