摘要
用所制备的复制型M13mp18和寡核苷酸(GACA)_4探针,分别与近交系AKR小鼠的9个个体及NOD小鼠的5个个体进行DNA杂交,结果提示,一种探针和近交系内各个体杂交的DNA指纹图完全一致;同一探针与不同近交系小鼠杂交的指纹图完全不同;不同探针与同一近交系小鼠杂交的指纹图也完全不同。说明用DNA指纹图分析,对建立各种实验动物标准的DNA指纹图,用于遗传监测、杂交育种、基因制图以及寻找遗传标记等均有实用价值和参考作用。
The purpose of this approach is preparing for two kinds of multilocus
probes棗bacteriophage M13mp18 and Oligonucleotide ( GACA ) 4, which are
of inexpensive and convenient.The preliminary studies of DNA fingerprints of two species of inbred strain mice using the two kinds of probes were carried out.
The DNA samples were isolated from nine individuals of inbred strain mouse AKR of parent, F1, F2 andF3 generations respectively, The DNAs were hybridized with labelled M13mp18 probe.The completely identical DNA fing- erprints of two kinds of inbred strain mice AKR and NOD were analyzed by oligonucleotide ( GACA ) 4. The DNA fingerprints of five individuals of inbred strain NOD were completely identical. But they were different from those of AKR strain. It has been demonstrated that patterns of DNA fingerprints are different in various inbred strain mice.The results are of important value in identifying different inbred strain animals, establishing standard DNA fingerprints of various laboratory animals, and in searching for genetic markers.
