湖南产尖吻蝮(Agkistrodon acutus)蛇毒类凝血酶组份的分离纯化及性质

本文研究了湖南产尖吻蝮蛇毒中的类凝血酶组份Ⅸ-I-I的分离,纯化及性质。该组份经聚丙烯酰胺凝胶电泳,十二烷基硫酸钠-聚丙烯酰胺凝胶电泳(SDS-PAGE),聚丙烯酰胺等电聚焦电泳,均为单一区带,分子量15100道尔顿,等电点4.5.组份Ⅸ-I-I 0.17mg/ml的纤维蛋白原凝固效应与人凝血酶5单位/ml接近。其血浆凝固活性是粗毒的10倍。精氨酸酯酶比活力20μmol/mg·min(底物TAME)。该组份不具有酪蛋白酶活性和纤维蛋白水解酶活性。

This paper studies the characteristics of the thrombin-like enzyme Fract. IX-I- separated and purified from Agkistrodorncutus venom from Hu Nan.Fract. IX-I-I possesed the clotting activity,Fract. IX-I-I 0.17mg/ml. is eqivalento the activity of human thrombin 5μ/ml The molecular weight is 15100 dolton as determined in SDS-PAGE. The isoelectric point is about4.5 by isoeletric focusing. The arginine esterase activity is about 20umol/mg min (substrate TAME). Fract. IX-I-I does not posses cascin and fibrin protelytic activity.
