摘要
25例哮喘患者一步法得到茶碱的群体药动学参数 Ke 0.0873±0.0224h^(-1),Vd0.5152±0.0968 L/kg,CL44.98ml/kg·h。当取一点稳态谷浓度作 Bayesian 反馈可得到比较准确的个体药动学参数:12例老年 COPD 患者 Ke0.0545±0.0107h^(-1),CL29.36±4.72ml/kg·h;18例哮喘患者 Ke0.0871±0.0299h^(-1),CL45.48±15.14ml/kg·h。两组患者反馈点实测血药浓度与预测血药浓度相关性均为 r>0.99(P<0.01)。由此可设计出合理的给药方案,并预报可信的血药浓度。
The population pharmacokinetic parameters for theophyl- line were obtained in 25 asthma patients using one stage method.These parameters were as follows:Ke 0.0878±0.0224/h.VD 0.5152±0.0968 L/kg, CL 44.98 ml/kg·h.Bayesian feedback method was used for predicting individual pharmacokinetic parameters.Bayesian analysis of one steady-state trough serum theophylline concentration showed accurate prediction: 12 aged COPD patients in group I,Ke 0.0545±0.0107/h,CL 29.36± 4.72ml/kg·h,18 asthma patients in group Ⅱ,Ke 0.0871±0.0299/h,CL 45.48±15.14 ml/kg·h.The correlation cofficients between predicted and observed serum concentration were both higher than 0.99(P<0.01) in these two groups.It indicated that population-base Bayesian analysis could de- vise rational dosage regimen and provide the credible prediction of theophy- lline serum concentration.
