摘要
用鼠抗人甲状腺球蛋白(TG)单克隆抗体(18A_1,IgG_1)免疫家兔,抗血清流穿正常小鼠 Ig 与无关单抗(MIg-Lp-1-32)亲和层析柱,吸除抗同种型、同种异型抗体。经 ELISA 竞争抑制、中和抑制试验证明制备出抗 TG 独特型抗体(ATG-Ab_2)。ATG-Ab_2与 TG 免疫小鼠的脾细胞体外作用18h,能特异地抑制小鼠脾细胞分泌抗 TG,而对经 Fn 免疫的小鼠脾细胞分泌抗 Fn 无影响,表明其具有下调节免疫的作用。
Mouse Monoclonal anti-thyroglobulin antibody (18A1 IgG1) was used to immunize the rabbits.The obtained antisera were made anti-Id specific by a series of adsorption on sepharose 4B immunoadsorbents coated with nomal mouse immunoglobulin (MIg) and anti-legionella pneumophila McAb (Lp-1-32 IgG1),which shares isotype,allotype with mAb used for immunization.The results of competitive inhibition ELISA and binding in- hibition ELISA indicated that the activity of anti-thyroglobulin anti-idiotypic antibody (ATG-Ab2) was obtained.Spleen cells of mouse immunized with TG or Fn were cultured with ATG-Ab2 for 18h.The results show that ATG -Ab2 could suppress the anti-thyroglobulin antibody secretion and could not suppress the anti-fibronectins antibody secretion.The results suggest that anti-idiotypic antibody may have an effect on down-regulation of auto- immunity.
