摘要 应用高效液相色谱法;研究分离测定茶儿茶素的结果表明,采用茶样沸水浴振荡浸提20分钟,SEP-PAK C_(18)小柱净化试样的前处理方法和μBondapak C_(18)不锈钢柱,二甲基甲酰胺、甲醇、醋酸和水4成分系统流动相,流量、浓度线性梯度洗脱,柱温30℃等色谱条件。能使儿茶素组分在25分钟内得到理想的分离。应用紫外检测器(280nm)检测各儿茶素,最低检出限在5.7×10^(-8)g以下,添加回收试验,回收率在85.13%—97.56%之间。5种儿茶素组分回归方程的相关系数为0.9995—0.9999,线性关系良好。与文献报道比较,分离度提高,分析时间缩短,稳定性,重现性好,灵敏度高,适于大量试样的分析。 The purpose of this studied is to establish a simple,convenient and rapidmethod for determining tea catechins with HPLC.Results showed that teacatechins could be well seperated in 25 minutes by extracting tea sample for20 minutes in boiling water,purifying sample by the pre-treating methodof using SEP-PAK C_(18),employing μBondpark C_(18)stainless column,usingdimethylformamide,methyl alcohol,acetic acid and water as flow phase,and linear gradient elution of flowrate and concentration,with the tempe-rature of column 30℃.The lowest detectable limits of catechins were below5.7×10^(-8)g by using uv detector(280nm).The rate of recovery varied from85.13% to 97.56%.The correlative coefficients of regressive equation offive kinds of catechins varied from 0.9995 to 0.9999,which was a well linearrelationship.Comparing with the results of previous studies,this methodwas of higher seperating degree,shorter analysing time,better stablity andrepeatability and higher sensitivity.So,it yeas suitable for analysing greatdeal of tea samples.
