摘要 A型激酶锚定蛋白属于调节蛋白家族,主要功能是将蛋白激酶A靶定于特定的亚细胞结构,引导酶接近底物,促进酶对底物的磷酸化或去磷酸化作用;并能整合其他信号分子形成多酶复合物,从时空上控制着由细胞膜到细胞核或其他亚细胞的信号转导。A型激酶锚定蛋白2通过靶定蛋白激酶A调控晶状体水通道蛋白-0的渗透性,参与晶状体水代谢。本文就A型激酶锚定蛋白的结构特点、参与细胞信号转导的功能机制和对水通道蛋白-0的调节作用等方面进行综述,探讨A型激酶锚定蛋白与水通道蛋白-0及晶状体透明性三者的关系。 A-kinase anchoring proteins(AKAP)are a family of scaffold proteins that anchor protein kinase A(PKA)to specific subcellular sites and guide enzyme close to the substrate,then promote phosphorylation or dephosphorylation.AKAP also regulate the spatiotemporal signaling from cell membrane to nuclear or other subcellular by providing a framework for the formation of multi enzyme complexes with other signal molecules.AKAP2 anchored PKA may modulate the permeability of aquaporin-0(AQP0)to preserve fluid circulation in the lens.This article reviews the A-kinase anchoring protein structural features and functional mechanisms involved in cellular signal transduction,and its regulation of AQPO.Then we discuss the relationship among AKAP,AQPO and lens transparency.
