摘要
以乌桕当年生幼嫩茎段为外植体,研究不同种类和不同浓度植物生长调节剂对其组织培养过程的影响,建立乌桕离体快繁再生体系。结果表明:芽体诱导培养以MS+NAA 0.2 mg/L+6-BA3.0 mg/L为最佳培养基,萌芽率最高,达87.19%;在启动培养中,为了控制褐化现象,在培养基中添加2.0 g/L PVP效果最好;增殖培养以MS+6-BA 1.0 mg/L+NAA 0.10 mg/L为最佳培养基;1/2 MS+IBA 1.0 mg/L+NAA 0.1 mg/L诱导出的生根率最高。
The fast in vitro propagation and regeneration systemof Sapium sebiferum was established by taking young stem segment of S.sebiferum as explants,and the influences of different types and concentrations of plant growth regulators on the progress of S.sebiferum tissue culture were examined.The results showed that the optimum medium formula for bud induction culture was MS +NAA 0.2 mg/L +6 -BA 3.0 mg/L,which resulted in the highest bud rate as 87.19%.At the initial culture stage,adding 2.0 g/L PVP to the medium could well control the browning phenomenon.The optimum culture medium for multiplication was MS +6 -BA 1.0 mg/L +NAA 0.1 mg/L.The highest rooting rate was induced by medium as 1 /2 MS +IBA 1.0 mg/L +NAA 0.1 mg/L.
