摘要
采用高效液相色谱-电喷雾质谱(HPLC-ESI-MS)法测定桑葚中花色苷含量及种类。Zorbax SB-C18色谱柱(250 mm×4.6 mm,5μm),流动相为乙腈-10%甲酸水溶液,流速1.0 mL/min,检测波长520 nm。以矢车菊素-3-葡萄糖苷(Cy-3-G)为标准,外标法定量;并通过离子碎片对花色苷组成进行定性分析。结果发现,每100克桑葚鲜果含花色苷(711.7±49.5)mg;分别为矢车菊素-3-(2G-葡糖芸香糖苷)、飞燕草素-3-芸香糖苷-5-葡萄糖苷、矢车菊素-3,5-二葡萄糖苷、矢车菊素-3-葡萄糖苷、矢车菊素-3-芸香糖苷、天竺葵素-3-葡萄糖苷、天竺葵素-3-芸香糖苷和飞燕草素-3-芸香糖苷等8种花色苷。
Investigation of the content and variety of anthocyanins in mulberry by high-performance liquid chromatography with electrospray ionization-mass spectrometry (HPLC-ESI-MS). The analysis was carried on Zorbax SB-C18 column (250 mm × 4.6 mm, 5μm). The mobile phase was acetonitrile and 10%formic acid-water at the flow rate of 1.0 mL/min, detection wavelength was 520 nm. Mulberry anthocyanins were quantitated by cyanidin-3-glucoside according to external reference method and identified on the basis of ion fragments. The results showed that about (711.7 ±49.5) mg anthocyanins in 100 g fresh mulberry; eight kinds of anthocyanins were found in mulberry, such as cyanidin-3-(2G-glucosylrutinoside), delphinidin-3-rutinoside-5-glucoside, cyanidin-3,5-diglucoside, cyanidin-3-glucoside, cyanidin-3-rutinoside, pelargonidin-3-glucoside, pelargonidin-3-rutinoside, and delphinidin-3-rutinoside.
