摘要
目的:建立正斜丸的质量控制方法。方法:采用薄层层析法鉴别正斜丸中黄芪、白附子、防风、秦艽,采用高效液相色谱法-蒸发光散射检测器法对正斜丸中黄芪甲苷含量进行测定。色谱柱为Kromosil C18柱(4.6mm×250mm,5μm),流动相为乙腈:水(34∶66),流速为1.0ml/min。蒸发光散射检测器参数为:飘逸管温度100.8℃,N2流速为2.7L/min。结果:用薄层色谱法可鉴别出正斜丸中黄芪、白附子、防风、秦艽的特征斑点,阴性对照无干扰,专属性强。黄芪甲苷在50.12~300.7μg/ml范围呈较好线性关系,r=0.9995,平均加样回收率为98.42%,RSD为1.09%(n=6)。结论:本文建立的方法准确可靠,重现性好,能有效控制正斜丸的质量。
Objective: To establish the quality method for Zhengxie pill. Methods: TLC was used for the identification and HPLC-ELSD was used for the determination of astragaloside glycoside content in the Zhengxie pills. Kromosil C18 column (4.6mm×250 mm, 5μm)was used;Acetonitrile:H2O (34:66) was used as a mobile phase;Flow rate was 1.0ml/min;Detection wavelength was 358nm. ELSD parameters:elegant tube temperature was 100.8℃, the flow rate of N2 was 2.7L/min. Results:The characteristic spots of astragalus, white monkshood, windbreak and gentiana could be identified, yet without interference of negative control. Astragaloside glycoside showed good linear relationship in 50.12~300.7μg/ml (r=0.9995), the average recovery was 98.42%, RSD was 1.09%(n=6). Conclusion:The method showed good precision and reproducibility, the result is accurate, which can be used for quality standard for Zhengxie pill.
