Current Status and Future Perspectives of Tissue Engineering Skin

摘要 如何及时有效的覆盖创面是烧伤、创伤引起的皮肤缺损治疗的关键,对于大面积皮肤缺损患者而言,自体皮源的缺乏是其救治的难点,组织工程皮肤移植为解决这一难题带来了新希望。本文就目前组织工程皮肤研究现状和临床应用情况作一综述,内容包含组织工程表皮、组织工程真皮和组织工程复合皮肤。同时对未来组织工程皮肤研究方向进行了概述,着重强调了干细胞、免疫豁免以及新技术、新材料的使用。未来研究的最终目标是能够在体外或体内构建出结构和功能均能接近正常皮肤的永久存活的组织工程皮肤。 Major challenge in the treatment of cutaneous defects following severe burn or trauma is how to close wounds rapidly so that dehydration and infection is prevented.In addition to autologous skin,tissue engineering skin(TES) grafts have been recognized as a promising alternative source of autografts.This article has reviewed current status of research in TES and its clinical application,including tissue engineering epidermis,tissue engineering dermal scaffold and composite skin.In addition,future perspectives of tissue engineering skin,in particular,those containing stem cells,with immune privilege,as well as emerging techniques and materials that are likely to become important in future studies have been reviewed as well.The ultimate goal of TES research is to establish a permanently survivable skin substitute that mimic normal human skin in both structure and functions.
